FLOW CYTOMETRY with TRITON
Wash cells with PBS
Trypsinize cells, collect in media, and spin down at 300g for 5 min
Resuspend in PBS and spin
Add 1 ul Zombie for 1 million cells
Incubate 15 mins in dark
Wash w/CyTOF buffer and spin
Add 4 mL 1.6% PFA in PBS and incubate for 15 mins
Add 1 mL 0.1% Triton in PBS and incubate for 20 mins
Wash twice at 600g for 5 mins
Add blocking and antibodies
Incubate overnight
Wash twice with CyTOF buffer
