Pseudomonas Aeruginosa PA14 Infection Assay - C. elegans

Streak Pseudomonas aeruginosa PA14 on to a plate of Brain heart fusion. This plate can be maintained for ~14 days. Stream fresh if the plate is more than 14 days old.

Grow the bacteria in Brain heart fusion broth overnight with no antibiotics.

Plate the bacteria on fresh NGM plates. Make sure to spread the bacteria throughout the plate (worms tend to evade PA-14, so don't leave empty spaces within the plate)

Grow the bacteria for ~24 hours. If the bacteria grows, the plate will look green in color.

Transfer worms on to the plate for day 1 (with fudr) 

Transfer worms to a fresh plate on day 2 (with fudr)

Transfer worms to a fresh plate on day 3 (with fudr)

Transfer worms to a fresh plate on day 5 (with fudr)

Transfer worms every other day until the assay/lifespan ends
