Dual-luciferase reporter and enzyme-linked immunosorbent assays

Cells were co-transfected with the indicated firefly luciferase reporter plasmid, pRL-TK-renilla luciferase plasmid, and TUT7 or empty control plasmid, and cultured for 48 h. Cells were harvested and lysed, and firefly and renilla luciferase activities were determined using the Dual-Luciferase reporter assay system (Promega, Madison, WI) according to the manufacturer’s instructions. The levels of cytokines in cultural supernatants were measured by DuoSet ELISA systems (R&D Systems, Minneapolis, MN) following the manufacturer’s instructions.
