U87MG cells seeded at 6.7 × 104 cells per well in a 12-well plate were transfected with 50 nM siGenome SMARTpool EGR1 (catalog number M-006526-01; Dharmacon) or AllStar negative-control small interfering RNA (siRNA; catalog number 1027280; Qiagen), using 1.33 μl of the Dharmacon DharmaFECT 1 transfection reagent (catalog number T-2001-02). At 24 h posttransfection, cells were infected with VEEV TrD (MOI, 5) for 1 h. After infection the medium was replaced with fresh medium. At 25 h after infection, supernatants or cells were collected for analysis.
