For evaluation of the system PSF, green fluorescence labelled 500 nm diameter nanobeads (PS-Speck™ Microscope Point Source Kit P7220 by Thermofisher) were used. To achieve a bead density sufficient for PSF evaluation, 0.5 μl of the stock solution were diluted by 1:10,000 in ProLong™ Glass Antifade (P36984, Fisher). The antifade mounting solution was chosen to match the refractive index of the sample holder and 3D-printed prism of 1.52, in an effort to reduce aberrations between the different media. The solution is distributed into custom 3D printed mini wells, sealed over with a coverslip and cured at room temperature for 18 h. The final test slide can be imaged in the same configuration as the fixed microscope slide samples.
All other samples are of commercial origin and bought in their imaged form.
