CEREP 80 panel assay was carried out by Eurofins Cerep Panlabs facility at France. The data is tabulated in Supplementary Table 4. A series of binding assays was carried out to understand the off-target toxicity of CA-170. CA-170 was tested at 10 μM test concentration in duplicates. Compound binding was calculated as a % inhibition of the binding of a radioactively labeled ligand specific for each target. In each experiment the respective reference compound was tested concurrently with CA-170, and the data were compared with historical values determined at Eurofins. The experiment was accepted in accordance with Eurofins validation standard operating procedure. The results are expressed as a percent of control specific binding; (measured specific binding / control specific binding) × 100 and as a percent inhibition of control specific binding; 100−(measured specific binding / control specific binding) × 100 obtained in the presence of CA-170. The IC50 values (concentration causing a half-maximal inhibition of control specific binding) and Hill coefficients (nH) were determined by non-linear regression analysis of the competition curves generated with mean replicate values using Hill equation curve fitting.
