The animal model of CIA was established according to the previously described protocol [17]. Male Sprague–Dawley rats (8 weeks old) were immunized with an emulsion composed of equal parts of Freund’s complete adjuvant (FCA) containing 4 mg/ml heat-killed Mycobacterium tuberculosis (Chondrex) and bovine type II collagen (CII) solubilized at 2 mg/ml in 0.05 M acetic acid. The rats received an intradermal injection of 200 μl of emulsion (200 μg of bovine CII) into the dorsum. Booster doses were administered on day 8, with subcutaneous 100-μl injections of the same emulsion into the base of the tail. The onset of CIA was between days 12 and 16 after the first immunization.
