The genomic DNA cleavage assays for gene knockouts were performed using GeneArt Genomic Cleavage Detection Kit (Invitrogen) according to the manufacturer’s protocol. Briefly, genomic DNA was extracted from 5 × 105 lentiCRISPRv2-knockout bulk cells. Primers were designed to amplify the specific Cas9/sgRNA genetically modified region by PCR. The primer sequences were listed in Supplementary Table 2. The modifications (the insertion, deletion, or mismatched DNA) of the interested region from the PCR products were then cleaved and detected by the Detection Enzyme from GeneArt Genomic Cleavage Detection Kit.
