Both cell lines (Molm14, MV4;11, U937, HL-60, and THP-1) and primary cells were cultured in RPMI-1640 medium (Gibco) supplemented with 10% Fetal Bovine Serum (Gemini) and 100 μg/ml penicillin/streptomycin (Life Technologies). Molm14-R (Molm14 cells resistant to 60 nM Lestaurtinib) were cultured in the above medium supplemented with 60 nM Lestaurtinib. BaF3 cells with FLT3 point mutations were generated as previous described.17 All cells were cultured at 37 °C with 5% CO2.
