Fresh leaf tissue (0.2 g) was collected for the measurement of anthocyanin content. In brief, anthocyanins were extracted in 10 ml acidified methanol (99 CH3OH:1HCl, v/v) at 4 °C for 24 h in the dark, clarified by centrifugation at 12,000 × g for 2 min, and the absorbance of supernatants was determined using a UV-visible spectrophotometer (UV-2550, Shimadzu, Japan) from optical density (OD) at 530 and 600 nm. Anthocyanin concentrations were expressed as U∙g-1, where U was calculated as (OD530–OD600)/0.1 [18].
