Cell surface affinity of TNB-486 to human CD19 was assessed using an ALL cell line expressing human CD19 (Nalm6). Antibodies bound per cell was determined as described earlier using Alexa Fluor 488-labeled TNB-486 (Alexa Fluor labeling kit by Thermo Fisher Scientific), and Bangs Lab Quantum™ Alexa Fluor® 488 was used for generating the standard curve. To calculate affinity of the molecule (KD), antibodies bound per cell was determined and used to further calculate free TNB-486 concentration using the formula: StartingTNB−486−BoundTNB−486=FreeTNB−486.
