Overlapping peptide libraries were designed on the basis of the GII.4 Farmington Hills strain (Genbank accession number AY502023). Peptides were 15 amino acids long and spanned each ORF, with neighboring peptides overlapping by 10 residues. All peptides were synthesized by GenScript (Piscataway, NJ), resuspended in DMSO at a concentration of 40 mg/mL, and stored at –80°C. Overlapping peptide pools containing up to 25 peptides were generated as shown in Figure 3. Pools were made in serum-free RPMI 1640 with each peptide at 0.8 μg/mL and stored at –80°C.
