Culture of Human Monocytic Cell Line THP-1

THP-1 (ATCC: TIB-202) is a human monocytic cell line derived from an acute monocytic leukemia patient. THP-1 cells were cultured in RPMI 1640-GlutaMAX (Gibco, Thermo Fisher Scientific) supplemented with 10% heat inactivated FBS and 1% P/S at 37°C under 5% CO₂. Cells were grown to a density of 1–8 × 10⁵ cells/ml and used for experiments between passage 2 and 10. Differentiation of THP1 cells into macrophage-like cells was induced by stimulation with 50 ng/ml phorbol-12-myristate-13-acetate (PMA) (Sigma) for 72 h. Following differentiation, PMA-containing media was replaced with fresh media, and cells were rested in culture for 24 h.
