Overnight cultures were inoculated from frozen stocks. 100 μl of each strain was spread on each plate in triplicate and allowed to dry by evaporation. At each time point, the number of viable cells was determined by “coring” a section of the lawn with a sterile Pasteur pipet (inner diameter ~ 1.5 mm), resuspending each core in 50 μl of LB, and titering by serial dilution and plating onto LB medium; the limit of detection is < 100 cfu/ml54,55.
