NucA-PTX-Rh and FAM-NucA-PTX-Rh at a single dose of 5 mg kg−1 were given to the tumor-bearing nude mice, respectively, by intravenous route (i.v.) via the tail vein. The mice were killed at 0.5, 1, 1.5, 2, 2.5, 3, 3.5, and 4 h. The tumors were then collected, chopped and homogenized. The fluorescence of rhoamine in NucA-PTX-Rh treated mouse tumor homogenates and the fluorescence of rhodamine and FAM in FAM-NucA-PTX-Rh treated mouse tumor homogenates were analyzed by a fluorescence plate reader.
