Cryopreserved tracheal washes and plasma were thawed at room temperature for 30-60 minutes and mixed well by pipetting up and down prior to loading. An aliquot of 5.5 μL of each sample was pipetted into each macrochambers of a CodePlex chip pre-patterned with a complete copy of a 23-plex antibody array. 2% BSA/PBS was used as background control. The chip was then loaded into an IsoLight automation system and various proteins were measured by fluorescence ELISA and analyzed by the IsoSpeak software using the IsoPlexis Human Adaptive Immune Panel: GM-CSF, Granzyme B, IFN-γ, IL-10, IL-13, IL-15, IL-17A, IL-2, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IP-10, MCP-1, MIP-1α, MIP-1β, Perforin, sCD137, TGF-β1, TNF-α, TNF-β.
