Determination of myeloperoxidase (MPO) activity

To quantify neutrophil infiltration, MPO activity in the homogenized lung tissues was evaluated by using a MPO Detection Kit (Nanjing Jiancheng Bioengineering Institute, Nanjing, China). Briefly, lung samples were homogenized and then centrifuged at 12,000g at 4 °C for 20 min. Subsequently, the supernatants were collected. MPO activity in supernatants was determined at 460 nm using a 96-well plate reader, and presented as units per gram of total protein (U/g). Total protein concentrations were calculated via the BCA protein assay kit.
