4.7. Sphere Assay in Methylcellulose

For quantification of anchorage-independent sphere-forming capacity, 0.5 × 103 single cells were seeded into 96-well ultralow attachment plates in MSC medium containing 1% methylcellulose (Sigma-Aldrich). Cells were cultured as described above. After 10 days, spheres with a diameter of over 50 μm were counted using a cell culture microscope.
