MTT (Sigma-Aldrich; Thermo Fisher Scientific, Inc.) was used to assess the viability of SW1990 and AsPC-1 cells. Cells (6x103 per well) were seeded in 96-well plates and incubated for 24 h at 37˚C. Following transfection, cells were cultured for another 0, 24, 48 or 72 h at 37˚C. Subsequently, MTT (5 mg/ml) was added into each well and incubated at 37˚C for 4 h. Then, 150 µl DMSO (Sigma-Aldrich; Merck KGaA) was added to solubilize formazan. The absorbance at 490 nm was measured by a Multiscan Spectrum (Beijing Putian New Bridge Technology Co., Ltd.).
