20S proteasome enzymatic activity analysis

To avoid affecting the enzymatic activity of the proteasome, 0.5% CHAPS buffer was used to prepare the total cellular protein lysate. The proteasome activity was determined in the total protein lysates using a Proteasome 20S Activity Assay kit (cat. no. ab112154; Abcam) according to the manufacturer's instructions. The fluorescence intensity was measured at the excitation/emission wavelengths of 490/525 nm using a VICTOR X2 fluorescent microplate reader (PerkinElmer, Inc.).
