Cell viability was determined by the tetrazolium salt 3-[4,5-dimethylthiazol-2-yl] -2,5-diphenyltetrazolium bromide (MTT; Sigma, St. Louis, MO, USA) assay [23]. Primary astrocytes were seeded in triplicates in 24-well plates at 1 × 105 per well and cultured for 7 days in the NB+ medium. Astrocytes were treated with different antidepressants at different concentrations for 24 h following overnight starvation. Thereafter, MTT was added into each well and incubated at 37 °C for 1 h. The resulting formazan crystals were dissolved in dimethyl sulfoxide (Sigma, St. Louis, MO, USA). Optical density was measured at 490 nm.
