On reaching 80–90% confluence, DPSCs expanded with or without H2O2 (0–200 μM), were treated with StemPro® Accutase (Thermo Fisher Scientific) and PD rates calculated from cell counts throughout their proliferative lifespans, as previously described [12, 38, 39]. Cumulative PDs were subsequently plotted against time in culture, with the onset of cellular senescence confirmed when DPSCs underwent < 0.5 PDs/week [12, 38].
