MDA-MB-231 (and SUM159 cells) were seeded into 96-well plates with black bottom (2500/well). Then, 24 h later, cells were treated with a dose range of celecoxib for 96 h and then incubated with PrestoBlue™ Cell Viability Reagent (Thermo Fisher Scientific) for 40 min at 37 °C/5% CO2. Fluorescence measurements (excitation 535 nm, emission 615 nm) were then taken on the 96-well plates and the fluorescence values were recorded and analyzed. Since the fluorescence values have a linear correlation with the cell numbers, the data were used to calculate the percentage of cell viability inhibition following celecoxib treatment.
