Fluorescein Diacetate (FDA) Staining

Fluorescein diacetate was used as a negative stain to identify vacuole patterns in root hairs. Plants were treated with a 0.05% w/v solution of FDA (Sigma) in ddH2O for 15–20 min and analyzed under the fluorescence microscope.
