Absorbance versus temperature profiles were obtained with a Cary 50 spectrophotometer equipped with a Peltier cell holder. Melting experiments were performed at 295 nm in 10 mM sodium cacodylate (pH 7.2) and 100 mM KCl. The heating/cooling rate was 0.5°C/min. The melting points were determined from derivative plots of the melting curves. The oligonucleotide concentration was 5 μM.
