Primary human naïve CD4+ were isolated from anonymous healthy donor peripheral blood (STEMCELL Technologies #19555). After naïve CD4+ T-cell isolation, Th1 and Th2 cells were differentiated using the supplier’s protocol (R&D cat. #CDK001 and #CDK002). Briefly, naïve CD4+ T cells were activated with Human T-cell activator CD3/CD28 dynabeads at a 1:2 cell:bead ratio and cultured for 24 h with Th1/Th2 differentiation media. One day after T-cell activation, primary human T cells were transduced with the methods mentioned above. During culture, T cells were cultured in T-cell differentiation media (Th1 or Th2 depending on the experiment) for at least 14 days.
