Uptake of [14C]-palmitic acids by tissue explants and cells was measured as described previously42 with slight modification. In short, tissue pieces or cells were incubated in HBS solution containing 0.1% fatty acid-free BSA and [14C]-palmitic acids (2 µM) at 37 °C for 15 min or on ice followed by washing four times with ice-cold HBS solution containing 0.1% fatty acid-free BSA. The tissue pieces and cells were then lysed in RIPA buffer and the supernatant was used for liquid scintillation counting. The small amount of extracellular [14C]-palmitate remaining was corrected from the [14C]-palmitate content in samples. Cellular [14C]-palmitate uptake was normalized to total protein content in each sample.
