Two × 107 BJAB cells were harvested after 48 hr in the presence of 100 ng/mL doxycycline or normal media, and washed twice with sterile PBS. Cells were then fractionated according to the QProteome Mitochondrial isolation kit (Qiagen). For ATP5G1 quantitation, resulting protein samples were heated for 7 min at 95°C and western blotted. For mass spectrometry, proteins were precipitated and resuspended in urea buffer for digestion.
