Microarray analysis was performed on RNA prepared from HCCC-9810 and RBE cells infected with PTEN adenovirus, using the GeneChip Human Genome U133 Plus 2.0 Array (Affymetrix) according to the manufacturer’s instructions. Data generated by the GeneChip Scanner 3000 were then transformed with Affymetrix GeneChip Command Console software (Affymetrix) and saved as .CEL files. Transformed data preprocessing (background correction, normalization, and expression calculations) was performed using Robust Multiarray Analysis. Significance analysis of microarrays was used to select genes that exhibited differential expression between samples, and the R language pack was used to calculate the significance of such differences (q value). A significant difference was defined as a fold change >2 and q < 0.01. GO analysis and pathway analysis were applied to analyze the main function of the differentially expressed genes according to the GO classification of NCBI and KEGG (Kyoto Encyclopedia of Genes and Genomes) database (52) (53).
