Groups of 45 mouse or 55 human islets were perifused at a flow rate of 0.5 mL/min with a physiological salt solution [31] supplemented with 2 mM or 20 mM glucose in the absence or presence of compounds of interest using a temperature-controlled perifusion system [30]. Perifusate fractions were collected at 2 min intervals and secreted insulin was quantified by radioimmunoassay [32]. SR141716A and AM251 were dissolved to 10 µM in DMSO, such that the final DMSO concentration was 0.1%, which was also used for control (vehicle) perifusions.
