Surface sensing of translation (SUnSET) assay was performed as previously described51. Briefly, cells were incubated with 10μg/mL puromycin (Thermo Fisher) for 10 min followed by washing with ice cold PBS and lysing with RIPA buffer. Cell lysates were loaded onto SDS–PAGE, and western blotting was performed with a mouse anti-puromycin monoclonal antibody (Millipore), and normalized against Ponceau S staining (Sigma).
