Four-d–old seedlings were incubated in MS liquid medium containing 2 μM of FM4-64 dye for 15 min and 30 min, followed by immersion in MS liquid medium for a few seconds before imaging under an LSM800 confocal laser-scanning microscope (Zeiss).
