To form the two groups involving preconception ethanol exposure compared to control, HuC:GFP and hcrt:EGFP adult females of approximately 6 months of age were trained to consume control-gelatin and 10% ethanol-gelatin as described above. Following 14 days of daily consumption for their morning meal, these animals were then bred with either ethanol-naïve HuC:GFP or ethanol-naïve hcrt:EGFP males, respectively, and their offspring were grouped together and grown to 28 hpf, 6 dpf and 12 dpf for the subsequent behavioral and live-imaging experiments.
