When the multinucleated osteoclasts were formed, the medium was aspirated, the cells were washed with phosphate-buffered saline (PBS), fixed with 4% paraformaldehyde at room temperature for 20 min, and stained for TRAP using a leukocyte acid phosphatase kit. After the staining, multinucleated osteoclasts which contained 3 or more nuclei, could be identified by counting the number of nuclei in an osteoclast using a Leica light microscope (Leica).
