Plasma membrane integrity was assessed by flow cytometry using Propidium Iodide (PI, Sigma-Aldrich) staining. PI was added to yeast cell suspensions to a final concentration of 5 μg/mL and incubated for 10 min at room temperature. Cells with red fluorescence [FL-3 channel (488/620 nm)] were considered to exhibit compromised plasma membrane integrity.
