8-week old C57BL/6 male mice were fed commercially-prepared 0.2% cuprizone chow (Envigo Co.) for either 6 weeks, leading to acute demyelination, or 12 weeks, leading to chronic demyelination and remyelination failure, as described in54. Following each treatment time point, half of the animals were allowed to recover for 3 weeks on normal chow (remyelination), with a minimum of 3 brains collected per time point. Littermates fed normal chow were used as controls, and sacrificed as appropriate (6, 9, 12 and 15 weeks) along with experimental animals to provide age-matched controls.
