Bone marrow was extracted from adult BALB/c mice and plated at a density of 2×10⁶ cells in culture medium. Granulocyte-macrophage colony stimulating factor (GM-CSF) and Interleukin-4 (IL-4) were added at 40 ng/mL and 20 ng/mL, respectively. Cells were incubated at 37°C and medium (including cytokines) was replenished after 3 days. On day 6, non-adherent and loosely-adherent cells were harvested by pipetting and re-suspended in culture medium.
