Transfection and luciferase reporter assay

HEK293 cells were cotransfected with ISRE promoter firefly luciferase reporter plasmid and a TK-Renilla luciferase reporter, together with vector or various DUBs, or control siRNA or siRNA targeting USP22. 24 h later, cells were infected with SeV for 8 h. Luciferase activity was measured with a Dual-Luciferase Reporter Assay System (Promega).
