MR-Egger regression is similar to IVW, except the intercept is not fixed to zero, and hence allows for horizontal pleiotropy. The intercept also captures the degree of horizontal pleiotropy, and a p<0.05 indicates the presence of such pleiotropy, which in turn suggests the IVW estimate may be invalid.34 MR-Egger requires the Instrument Strength Independent of Direct Effect assumption and has lower statistical power.
