Kasumi-1 or OCI-AML3 CD34+/- subsets were seeded (4×104 cells/200 μl) in triplicates in the presence of Ara-C (0–1.6 μM and 0–12.8 μM, respectively) or DNR (0–0.8 μM for both cell lines). After 48 hours, 20 μl/well of alamarBlue reagent (Bio-Rad Laboratories, Inc.) were added and after additional 18 hours, viable cells were measured using fluorometer (FLUOstar Galaxy, BMG Labtechnologies, Germany). The mean number of viable cells at varying concentrations of the drugs was normalized to the number of untreated (no-drug) cells and percentages were calculated.
