Prostate tumors were dissected from Ptenpc−/−Smad4pc−/−Trp53c−/−mTmG mice and washed in PBS. Tumors were then minced with a blade and dissociated to single cells following the MACS dissociation kit protocol (Miltenyi Biotec 130-096-730). Primary growth medium consisted of DMEM (Life Tech 11995073) with 10% FBS (Life Tech 10082147) and 1× Penicillin-Streptomycin (Life Tech 15140163). GFP-positive cells were sorted and incubated at 37°C in 5% CO2. The resulting primary murine prostate cancer cell line was named DX1.
