For assessment of meiotic progression, cells that had been incubated in sporulation medium for 36 h were fixed with ethanol, stained with DAPI, and photographed under wet mount conditions using a Nikon Optiphot equipped for epifluorescence as previously described (8). Sporulation efficiency was analyzed by phase-contrast microscopy.
