CD4+ T cells were purified from spleens and lymph nodes of OT-II mice and the cells were labelled with CFSE. The labelled cells (1×105 per well) were co-cultured with dendritic cells in the absence or presence of OVA323-339 peptide for 3 days in 96-well microplates. CFSE dilution was assayed by flow cytometry.
