4.2. Conjugation to BCA DOTA and Radiolabeling of h8C3 mAb

DOTA conjugation to h8C3 was performed in a similar manner to our previous work with this antibody with a minor alteration [13]. A 10-fold molar excess of DOTA-SCN was used in place of a 10-fold excess of CHXA’’-SCN. ²²⁵Ac labeling of DOTA-conjugated h8C3 was performed in an analogous method to that in Dawicki et al. [37]. All radiolabeling yields were greater than 99%. ¹⁷⁷Lu labeling was performed in a similar manner using a 10:1 µCi: µg ratio of isotope to DOTA conjugated mAb.
