4.10. 3H-Thymidine Incorporation Assay

The MSCs were cultured in 24-well culture plates at a density of 5 × 10⁵ cells/500 μL in SD condition for 6, 18, and 24 h. ApoA-I was added to the test samples at a concentration of 20 μg/mL. 3H-thymidine (2.0 µCi/mL) was added 2 h before cell harvesting. The radioactivity of the samples was measured on a Scintillation System LS 6500 (Beckman Coulter, Brea, CA, USA). Speed of label inclusion was calculated in pulses of 1 min per 10⁶ cells.
