Fresh kidney tissues were mounted using Tissue-Tek O.C.T. Unfixed frozen samples were cut into 5 μm thick sections and placed on glass slides. DHE (dihydroethidium, 10 μmol/L) was applied to each tissue section, and then, the slides were incubated in a light-protected humidified chamber at 37°C for 30 min. The resulting color reaction was immediately measured with a fluorescence microscopy (Leica, Germany) and analyzed by image densitometry using ImageJ software (NIH).
