RNA Extraction and RNA-Seq Library Construction

This experiment was divided into in two groups including Aconitine group and Aconitine/AS-IV group, in order to obtain the DEGs of AS-IV on aconitine-induced cardiac damage. The effect of aconitine on gene expression was eliminated accordingly.

Total RNA extraction was performed with TRIZOL reagents from Invitrogen following the manufacturer's instructions. RNA library construction was then performed by BGI Co., Ltd, Shenzhen, China (http://www.genomics.cn/). An Agilent 2100 Bioanalyzer was used to detect RNA concentration, RNA Integrity Number (RIN) value, 28S/18S, and fragment size to determine the integrity of RNA, and the purity of RNA as detected using an ultraviolet spectrophotometer NanoDrop (OD260/280) (Qian et al., 2014).
