2.8. 20S Proteasome Activity Assay

Measurement of 20S proteasome activity (relative to control) in MH-S cell lysates was determined using a 20S Proteasome Assay Kit (Cayman Chemical, Ann Arbor, MI, USA). Adherent cells (5 × 105) were treated in 12-well, polystyrene plates and lysates were collected after 20 h culture. Fluorescence resulting from SUC-LLVY-AMC substrate proteolysis was used to calculate the catalytic activity of the proteasome in cells following treatment with CSE, H2O2, or bortezomib.
