MTT (3-[4,5-dimethylthazol-2-yl]-2,5-diphenyltetrazolium bromide) Assay

The control or transfected SPC-A1 cells were seeded in 96-well plates at a density of 2000 cells/well (200 μL). After culture for 0, 6, 12, 24, 48, and 96 hours, cell viability was measured by the MTT procedure. Twenty microliters of MTT (5 mg/mL) was added to each well, and the plates were then continuously incubated for 4 hours. Reactions were terminated with 200 μL Dimethyl Sulphoxide (DMSO) for 5 minutes. The optical density value was measured at 490 nm wavelength by an ELx800 Automated Microplate Reader (Biotech, USA).
