Measurement of AMP:ATP ratio by high performance liquid chromatography (HPLC)

The amounts of AMP and ATP were quantified using an HPLC method essentially as described previously.46 Briefly, 2 × 106 cells were plated in 100-mm2 dishes and treated with 4OHT for 24 h. Cells were harvested in 0.2 N perchloric acid and neutralized with KOH. Nucleotides in cell lysates were analyzed by Thermo Ultimate 3000 HPLC (Thermo Scientific, Wilmington, DE, USA) using an Inno C18 column (Innopia, 05AL 02015).
